18. ĐÁNH GIÁ KHẢ NĂNG TĂNG SINH CỦA TẾ BÀO CAR-T KHI ĐỒNG NUÔI CẤY VỚI CÁC DÒNG TẾ BÀO UNG THƯ CD19 IN VITRO

Tóm tắt

Mục tiêu: Đánh giá khả năng tăng sinh của CAR-T khi đồng nuôi cấy với các dòng tế bào biểu hiện CD19+ in vitro.

Đối tượng và phương pháp: Chuyển nạp gen vào tế bào K562 sử dụng hệ véc tơ Transposon-transposase kết hợp với xung điện. Tách tế bào PBMC và phân tích PBMC bằng flow cytometry. Chuyển nạp vào PBMC sử dụng dung dịch chuyển nạp P3 Primary Cells, 1 µg plasmid SB100X và 3 µg plasmid CD19RCD137/pSB. Đồng nuôi cấy CAR-T và tế bào CD19+ theo tỉ lệ 1:1.

Kết quả: Trong 4 ngày nuôi cấy, CAR-T CD19RCD137 và CAR-T iCasp9-IL15 đều có khả năng tăng sinh khi đồng nuôi cấy với các tế bào CD19+. Tuy nhiên, số lượng CAR-T CD19RCD137 có xu hướng giảm dần, nhưng CAR-T thế hệ 4 vẫn tăng về số lượng khi nuôi cấy với K562 không biểu hiện CD19. Ở ngày nuôi cấy 11, gần như không còn tế bào CAR-T CD19RCD137 và suy giảm đáng kể lượng tế bào CAR-T iCasp9-IL15.

Kết luận: Tế bào CAR-T có khả năng tăng sinh mạnh khi đồng nuôi cấy với các dòng tế bào CD19+ nhưng không có khả năng tăng sinh khi nuôi cấy cùng các dòng tế bào CD19.
