Những người mang đột biến (ĐB) gen BRCA1/2 có nguy cơ mắc ung thư (UT) vú và buồng
trứng (BT) cao hơn. Tỷ lệ lưu hành các ĐB BRCA1/2 thay đổi theo chủng tộc và dân tộc. Tỷ lệ và
các nguy cơ liên quan đến các ĐB BRCA1/2 chưa được rõ ở người Việt Nam.
Mục tiêu: Xác định các ĐB BRCA1/2 cho bệnh nhân (BN) UT vú, UTBT có các yếu tố hội
chứng UT vú- UTBT di truyền (HBOC), từ đó, đề xuất cá nhân hóa đánh giá nguy cơ mang ĐB,
nguy cơ mắc UT vú, UTBT và kế hoạch sàng lọc, dự phòng.
Đối tượng: 33 BN được chẩn đoán UT vú và/hoặc UTBT mắc hội chứng UT vú- UTBT di
truyền (HBOC)
Phương pháp: Mô tả cắt ngang. Mẫu máu của các BN được thu thập và phân tích bằng Giải
trình tự thế hệ mới. Các ĐB được kiểm định bằng giải trình tự Sanger. Xác định các ĐB ở người
nhà của BN mang ĐB.
Kết quả: Xác định được 11 loại ĐB ở cả BRCA1 (9 BN) và BRCA2 (3 BN), trong đó
BRCA2:p.Ser1341Ter là ĐB mới, chưa từng được công bố. Bảy trong 19 người thân BN có ĐB
BRCA1/2. Tất cả đối tượng đều được tư vấn về khả năng mắc UT cũng như các biện pháp tầm
soát và dự phòng.
Kết luận: Nghiên cứu cho thấy ý nghĩa của cá nhân hóa xét nghiệm gen BRCA1/2 ở BN UT
vú, UTBT và sàng lọc di truyền ở người thân.
