31. NGHIÊN CỨU ĐỘT BIẾN GEN Pvcrt LIÊN QUAN ĐẾN KHÁNG CHLOROQUINE Ở PLASMODIUM VIVAX TẠI MƯỜNG TÈ, LAI CHÂU BẰNG REAL-TIME PCR VÀ GIẢI TRÌNH TỰ TOÀN BỘ HỆ GEN

Tóm tắt

Mục tiêu: Nghiên cứu nhằm đánh giá mức độ biểu hiện và đột biến gen Pvcrt liên quan đến kháng Chloroquine ở Plasmodium vivax tại huyện Mường Tè, tỉnh Lai Châu, Việt Nam.

Phương pháp: Nghiên cứu mô tả cắt ngang trên 48 mẫu máu dương tính P. vivax, thu thập từ tháng 10/2022-3/2024. Biểu hiện Pvcrt được định lượng bằng qPCR, đột biến được xác định qua giải trình tự toàn bộ hệ gen Illumina. Bổ sung tổng quan tài liệu quốc tế từ 2008-2023 về gen Pvcrt và kháng Chloroquine.

Kết quả: 14,6% mẫu có biểu hiện Pvcrt tăng cao, 4,2% mang đột biến C350T, 12,5% có chèn codon 10 (K10). Mối liên hệ giữa C350T và tăng biểu hiện Pvcrt có ý nghĩa thống kê (p < 0,05). Tổng quan tài liệu khẳng định vai trò của Pvcrt trong kháng thuốc tại nhiều khu vực Đông Nam Á và Nam Á.

Kết luận: Pvcrt là chỉ dấu tiềm năng trong giám sát kháng Chloroquine ở P. vivax. Khuyến nghị phân tích biểu hiện và đột biến Pvcrt vào chương trình giám sát sốt rét tại Việt Nam.
