THE PERFORMANCE OF THE REAL-TIME PCR ASSAY DETECTING PENA 60.001 IN THE DIAGNOSIS OF CEFTRIAXONE‑RESISTANT NEISSERIA GONORRHOEAE.

Abstract

Objectives: To evaluate the performance of a real-time PCR assay in detecting ceftriaxone-resistant Neisseria gonorrhoeae. Methods: A cross-sectional descriptive study was conducted using real-time PCR to detect the penA 60.001 allele on 131 clinical isolates of N. gonorrhoeae and 89 N. gonorrhoeae-positive urethral discharge specimens at the Microbiology Unit, Microbiology and Parasitology Department, Hanoi Medical University Hospital. Results: Among the 131 isolates from genital discharge, 61 (46.56%) exhibited a ceftriaxone-resistant phenotype. For these isolates, the real-time PCR assay for penA 60.001 showed a sensitivity and specificity of 60.66% and 94.29%, respectively. When applied directly to N. gonorrhoeae-positive clinical specimens, the assay demonstrated a sensitivity of 24.14% and a specificity of 100%. Conclusions: This real-time PCR assay may provide supportive evidence for the detection of ceftriaxone-resistant N. gonorrhoeae strains. Further optimizations are required to enhance the assay's sensitivity. Additional research is also needed to evaluate the efficacy of this method when using DNA directly extracted from clinical specimens.
