Objective: This study investigates Pvcrt gene expression and mutations associated with Chloroquine resistance in Plasmodium vivax isolates from Muong Te district, Lai Chau province, Vietnam.
Methods: A cross-sectional study was conducted on 48 blood samples positive for P. vivax, collected from October 2022 to March 2024. Pvcrt expression was quantified by qPCR, and mutations were identified using whole genom Illumina sequencing. In addition, a literature review (2008-2023) was performed to evaluate Pvcrt roles globally.
Results: 14.6% of samples showed increased Pvcrt expression. C350T mutation was present in 4.2% and codon 10 insertions (K10) in 12.5%. C350T was significantly associated with overexpression (p < 0.05). Literature confirms Pvcrt involvement in Chloroquine resistance across Southeast and South Asia.
Conclusion: Pvcrt plays a potential role in Chloroquine resistance surveillance in P. vivax. Integration of molecular monitoring of Pvcrt into Vietnam’s malaria control strategies is recommended.
